AOD‑9604 is a synthetic 15‑amino‑acid polypeptide derived from the C‑terminal segment (residues 177–191) of human growth hormone (hGH). Its primary function is to mimic the lipolytic (fat‑breaking) activity of growth hormone while avoiding side effects associated with full‑length hGH, such as proliferative effects and hyperglycemia. AOD‑9604 stimulates lipolysis—i.e., the breakdown of fat—and inhibits lipogenesis, thereby preventing the conversion of non‑lipid substrates into body fat. Unlike full‑length growth hormone, AOD‑9604 does not target the classical growth hormone receptor. Its lipolytic effect is thought to be mediated primarily via the β3‑adrenergic receptor (β3‑AR) pathway. Chronic administration upregulates β3‑AR RNA expression in adipocytes, and in β3‑AR knockout mice, its weight‑reducing effect is abolished. Moreover, because it does not act through the IGF‑1 pathway, it has no adverse impact on carbohydrate metabolism.
